Custom CTAG fusions domestication using the former GB Syntax

Category CDS1+CDS2 (B3-B4) domestication:

  • Upload your sequence to the “Domestication” tool. Please, note that since you are going to perform a custom domestication your input sequence should NOT include the initial ATG.
  • Leave the “Choose a category to domesticate to” field empty.
  • Fill “Custom prefix” = AATG and “Custom suffix” = GCAG (click here for a comparison between the old and new syntax)

Category CTAG (B5) domestication:

  • Add nucleotides GG to the 5’end of your sequence (you need to add two bases in order to keep the ORF). Keep in mind that your sequence should include a STOP CODON at the 3'end.
  • Upload your sequence to the “Domestication” tool.
  • Leave the “Choose a category to domesticate to” field empty.
  • Fill “Custom prefix” = GCAG and “Custom suffix” = GCTT (click here for a comparison between the old and new syntax)

- To add your domesticated parts to the GB database select "Type": OTHER (note that you cannot add your parts as standard parts since they do not adapt to the standard grammar).
